Selank is built from tuftsin, a four-residue immune-active fragment, extended by a three-residue tail to give the sequence Thr-Lys-Pro-Arg-Pro-Gly-Pro.

The extension is the entire design. Tuftsin on its own is cleaved rapidly; adding residues at one end slows that process without altering the original four. It is a clean illustration of a general strategy in peptide chemistry, where stability is engineered by addition rather than substitution.

Because the parent fragment is itself well characterised, comparisons between the two are unusually informative: any difference in behaviour can be traced to the tail rather than to an unknown structural change.
