Both changes serve one purpose. Native IGF-1 circulates bound to binding proteins that limit its availability, and the substitution at position 3 sharply reduces that binding. The molecule is essentially designed to escape its own regulatory system.

That property is exactly why it appears in cell-culture work: a growth factor that is not sequestered by binding proteins behaves far more predictably in a defined medium than the native form.
